结直肠癌作为全球高发的恶性肿瘤,其病理进程涉及复杂的分子调控网络。环状 RNA(circRNA)作为新 型非编码 RNA 分子,通过 miRNA 海绵、蛋白互作及翻译功能参与结直肠癌的发生发展,为肿瘤治疗提供了新 的靶点。该文系统解析了 circRNA 在结直肠癌中的生物合成、功能特性及失调机制,重点综述中医药调控 circRNA 治疗结直肠癌的研究进展;探讨中药复方、单味中药及中药单体成分通过调控 circRNA 网络干预结直 肠癌的分子互作模式,提出“中药成分-circRNA-靶蛋白”三维互作模型;指出结合单细胞测序与化学生物探 针技术解析动态调控网络的研究方向。该研究为开发基于 circRNA 靶点的中医药精准治疗方案提供了理论依 据,有助于推动传统医学与现代分子靶向治疗的深度融合。

Colorectal cancer,a highly prevalent malignant tumor worldwide,involves complex molecular regulatory networks in its pathological progression. Circular RNAs (circRNA),as novel non-coding RNA molecules,participate in the pathogenesis and progression of colorectal cancer through miRNA sponging, protein interactions, and translational functions, thereby providing new targets for cancer therapy. This article systematically elucidates the biogenesis,functional characteristics,and dysregulation mechanisms of circRNAs in colorectal cancer,with a focused review on the research advances in traditional Chinese medicine (TCM) regulating circRNA for colorectal cancer treatment. It explores the molecular interaction patterns by which TCM compounds, single herbs, and active components intervene in colorectal cancer through the regulation of circRNA networks, and proposes a three- dimensional interaction model of “TCM component-circRNA-target protein”. The review also highlights future research directions integrating single-cell sequencing and chemical biology probes to decipher dynamic regulatory networks. This work provides a theoretical basis for developing precision TCM therapeutic strategies targeting circRNA,contributing to the deep integration of traditional medicine and modern molecular targeted therapies.
