目的 通过多指标综合加权评分结合响应面法优化六经头痛复方的提取工艺。方法 采用层次分析法 (AHP)-熵权法(EWM)结合 Box-Behnken 响应面试验,分别以桉油精、甲基丁香酚、藁本内酯的含量与挥发油 提取率的综合评分为考察指标,筛选六经头痛复方中挥发油提取工艺;以 3’-羟基葛根素、葛根素、大豆苷、 阿魏酸、特女贞苷、木兰脂素成分的综合评分为指标,筛选六经头痛复方的水提工艺;并结合二者的工艺来综 合优化六经头痛复方的提取工艺。结果 六经头痛复方提取工艺为细辛、辛夷、川芎、藁本浸泡时间 0.5 h, 料液比 10∶1,提取时间 4 h,收集挥发油,保留水提液,滤渣与复方中剩余药味加 8 倍水,提取 2 h,提取 2 次,滤液与提取挥发油后的水提液合并。结论 采用 AHP-EWM 结合 Box-Behnken 设计响应面法优化六经 头痛复方的提取工艺方法可行,可为六经头痛复方新制剂的研发奠定基础。

[Abstract]
Objective To optimize the extraction process of Liujing Toutong Compound using multi-index comprehensive weighted scoring combined with response surface methodology. Methods The extraction process of volatile oils from Liujing Toutong Compound was screened using the Analytic Hierarchy Process (AHP)-Entropy Weight Method (EWM) combined with Box-Behnken response surface experiments. The comprehensive score of eucalyptol, methyleugenol, and ligustilide content, as well as the extraction rate of volatile oils, was used as the evaluation index. The water extraction process of Liujing Toutong Compound was screened using the comprehensive score of 3’-hydroxypuerarin,puerarin,daidzin,ferulic acid,specnuezhenide,and magnolin as the evaluation index. The extraction process of Liujing Toutong Compound was comprehensively optimized by combining the two processes. Results The optimized extraction process for Liujing Toutong Compound was as follows: Asari Radix et Rhizoma, Magnoliae Flos,Chuanxiong Rhizoma,Ligustici Rhizoma were soaked for 0.5 hours with a solid-to-liquid ratio of 10∶1, followed by extraction for four hours to collect volatile oils and retain the water extract. The residue was combined with the remaining herbs in the formula,extracted twice with eight times the volume of water for two hours each time,and the filtrate was combined with the water extract obtained after volatile oil extraction. Conclusion The use of AHP-EWM combined with Box-Behnken response surface methodology to optimize the extraction process of Liujing Toutong Compound is feasible and provides a foundation for the development of new formulations of Liujing Toutong Compound.
