目的 建立毛慈菇指纹图谱,结合化学计量学方法及质量标志物含量测定方法综合评价毛慈菇的质量。 方法 采用高效液相色谱(HPLC)法建立毛慈菇指纹图谱,采用聚类分析、主成分分析和正交偏最小二乘法-判 别分析(OPLS-DA)方法筛选毛慈菇的质量标志物,并对酪氨酸进行含量测定。结果 25 批毛慈菇 HPLC 指纹 图谱确定了 9 个共有峰,经对照品匹配指认出酪氨酸、香草醇 4-O-β-D 吡喃葡萄糖苷、blestriarene C、2,2', 7-三羟基-4,4',7'-三甲氧基-1,1'-联苯 4 个色谱峰。聚类分析及主成分分析均将 25 批毛慈菇样品分为 2 类:红黑子样品(M1~M8)为一类;白子样品(M9~M25)为一类。OPLS-DA 发现 7 号峰(blestriarene C)、 1 号峰(酪氨酸)、8 号峰、6 号峰、9 号峰(2,2',7-三羟基-4,4',7'-三甲氧基-1,1'-联苯)对区分不同外观性状 和不同产地毛慈菇的贡献较大,结合毛慈菇及其混伪品的成分差异结果,筛选出酪氨酸为毛慈菇的质量标志 物。25 批毛慈菇中酪氨酸的含量为 0.05%~0.42%,13 批毛慈菇混伪品酪氨酸含量均<0.01%。结论 该研究 建立的指纹图谱结合化学计量学方法及质量标志物含量测定方法准确、可靠,能够较客观、全面地评价毛慈菇 的质量。

[Abstract]
Objective To comprehensively evaluate the quality of Cremastra appendiculata by establishing its fingerprint,combined with chemometric methods and the determination of quality markers. Methods The fingerprint of Cremastra appendiculata was established using high-performance liquid chromatography (HPLC). Cluster analysis, principal component analysis,and orthogonal partial least squares-discriminant analysis (OPLS-DA) were employed to screen quality markers,and the content of tyrosine was determined. Results The HPLC fingerprint of 25 batches of Cremastra appendiculata identified nine common peaks. Through comparison with reference standards, four chromatographic peaks were identified as tyrosine,vanillyl alcohol 4-O-β-D-glucopyranoside,blestriarene C,and 2, 2',7-trihydroxy-4,4',7'-trimethoxy-1,1'-biphenyl. Both cluster analysis and principal component analysis classified the 25 batches into two categories:red-black seed samples (M1-M8) as one group and white seed samples (M9- M25) as another. OPLS-DA revealed that peak 7 (blestriarene C),peak 1 (tyrosine),peak 8,peak 6,and peak 9 (2,2',7-trihydroxy-4,4',7'-trimethoxy-1, 1'-biphenyl) contributed significantly to distinguishing Cremastra appendiculata with different morphological traits and origins. Combined with the compositional differences between Cremastra appendiculata and its adulterants,tyrosine was ultimately selected as the quality marker. The tyrosine content in the 25 batches of Cremastra appendiculata ranged from 0.05% to 0.42%,while the tyrosine content in 13 batches of adulterants was all <0.01%. Conclusion The fingerprint combined with chemometric methods and quality marker determination established in this study is accurate and reliable,providing an objective and comprehensive approach to evaluating the quality of Cremastra appendiculata.
