[摘要]
目的 研究平调颗粒的化学成分及大鼠灌胃给药平调颗粒后大鼠血清中的入血成分。方法 采用超高效 液相色谱-静电场轨道阱高分辨质谱(UHPLC-Orbitrap-MS)分析方法,根据保留时间、精确分子量及二级质谱 碎片并结合文献数据和对照品,对平调颗粒的化学成分及入血成分进行分析鉴定。结果 从平调颗粒中分析鉴 定出50种化学成分,主要为生物碱、有机酸、黄酮和皂苷类成分;在灌胃给药后的大鼠血清中分析鉴定出32种 入血成分,其中天麻 5种,钩藤 12种,杜仲 5种,白芍 5种,桑寄生 5种。结论 UHPLC-Orbitrap-MS 方法可 快速、高效、准确地分析平调颗粒的化学成分及入血成分,能为阐明平调颗粒的药效物质基础及其作用机制提 供参考依据。

[Abstract]
Objective To study the chemical components of Pingtiao Granules and their active components absorbed into the rat blood after oral administration of Pingtiao Granules. Methods An ultra-high performance liquid chromatography-electrostatic field orbital trap high-resolution mass spectrometry (UHPLC-Orbitrap-MS) was developed and used for the analysis. According to the retention time, accurate molecular weight, and fragments of multistage mass spectrometry,as well as the literatures and reference substances,the chemical compositions and its active components absorbed into blood of Pingtiao Granules were identified. Results A total of 50 chemical compositions from Pingtiao Granules were identified, and they are alkaloids, organic acids, flavonoids, and saponins. There were 32 active components in rat serum after intragastric administration of Pingtiao Graunles. Among them, 5 from Gastrodiae Rhizoma, 12 from Uncariae Ramulus Cum Uncis, 5 from Eucommiae Cortex, 5 from Paeoniae Radix Alba,and 5 from Talxilli Herba. Conclusion The chemical components of Pingtiao Granules and their active components absorbed into blood were analyzed by UHPLC-Orbitrap-MS method,which is rapid,efficient,and accurate. It may provide a basis for study the pharmacodynamic substances and effect mechanism of Pingtiao Granules.
