目的 比较产地趁鲜切制与传统加工防风饮片对小鼠的解热、镇痛作用,并对解热作用机制进行探讨。 方法 以产地趁鲜切制及传统加工防风饮片水提液(1.7 g·kg-1)进行药物干预,分别采用小鼠热板法镇痛实验及 干酵母致小鼠发热实验考察其解热、镇痛作用,测定并记录小鼠痛阈值及体温变化情况。采用 ELISA 法检测 小鼠血清白细胞介素 1β(IL-1β)、肿瘤坏死因子 α(TNF-α)、一氧化氮(NO)水平;Western Blot法检测下丘脑 组织中 Toll样受体4(TLR4)、IL-6、核转录因子(NF-κB)、磷酸化核转录因子(p-NF-κB)、环氧化酶2(COX2)蛋 白表达水平。结果 (1)与空白组比较,给药组小鼠的痛阈值均显著升高(P<0.05,P<0.01),均表现出一定 的镇痛效果,且防风传统加工组与趁鲜切制组小鼠表现出相似的镇痛效果,差异无统计学意义(P>0.05)。 (2)与空白组比较,其他各组小鼠在皮下注射20%干酵母混悬液6 h后,体温均显著升高(P<0.01),表明发热 模型复制成功。与模型组比较,各给药组小鼠的体温明显降低(P<0.05,P<0.01),均表现出一定的解热效果, 且防风传统加工组与趁鲜切制组小鼠表现出相似的解热效果,差异无统计学意义(P>0.05)。(3)与空白组比较, 模型组小鼠的血清TNF-α、NO、IL-1β含量均显著升高(P<0.05,P<0.01),下丘脑组织中 TLR4、p-NF-κB/ NF-κB、IL-6、COX2 蛋白表达均显著上调(P<0.01)。与模型组比较,各给药组小鼠的血清 TNF-α、NO、 IL-1β含量均显著降低(P<0.05,P<0.01),下丘脑组织中 TLR4、p-NF-κB/NF-κB、IL-6、COX2蛋白表达均 明显下调(P<0.05),而防风传统加工组与趁鲜切制组之间的差异无统计学意义(P>0.05)。结论 防风趁鲜切 制与传统加工饮片的解热、镇痛效果相似,解热作用可能与其降低炎症细胞因子表达有关。防风产地趁鲜切制 技术的实施具有药理学依据。

[Abstract]
Objective To compare the antipyretic and analgesic effects of fresh-cut and traditional processed slice of Saposhnikoviae Radix on mice,and to explore its antipyretic mechanism. Methods The aqueous extract(1.7 g·kg-1)of fresh-cut and traditional processed slice of Saposhnikoviae Radix from the producing area were used for intervention. Hot plate method and dry yeast-induced fever model were used to investigate the antipyretic and analgesic effects,then the changes of pain threshold and body temperature in mice were observed. ELISA was applied to detect the levels of serum interleukin-1 β (IL-1β), tumor necrosis factor alpha (TNF- α), and nitric oxide (NO)in mice. The protein expression levels of Toll like receptor 4(TLR4), IL-6, nuclear transcription factor(NF- κB), phosphorylated nuclear transcription factor(p-NF- κB),and cyclooxygenase-2(COX2)in hypothalamic tissue were determined by Western Blot. Results (1)Compared with the blank group, the pain threshold of mice in the treatment group was obviously increased (P<0.05, P<0.01). The administration group showed certain and similar analgesic effects. However,no significant difference was noticed(P>0.05). (2)Compared with the blank group,the body temperature of other groups of mice was significantly increased after subcutaneous injection of 20% dry yeast suspension for 6 h(P< 0.01),which indicated that the fever model was replicated successfully. Compared with the model group,the body temperature in the treatment group was significantly decreased(P<0.05,P<0.01). The administration group showed certain and similar antipyretic effects. However,no significant difference was found(P>0.05).(3)Compared with the blank group,the content of serum TNF-α,NO and IL-1β of mice in model group were significantly increased(P< 0.05,P<0.01). The protein expression levels of TLR4,p-NF-κB/NF-κB,IL-6 and COX2 in hypothalamic tissue were significantly up-regulated(P<0.01). Compared with the model group,the content of serum TNF-α,NO and IL-1β of mice in treatment group were significantly decreased(P<0.05,P<0.01). The protein expression levels of TLR4, p-NF-κB/NF-κB,IL-6 and COX2 in hypothalamic tissue were significantly down-regulated(P<0.05). There was no statistic difference between fresh-cut group and traditional processed group(P>0.05). Conclusion Similar antipyretic and analgesic effects were shown in fresh-cut and traditional processed slice of Saposhnikoviae Radix. The antipyretic effect may be related to reducing the expression of inflammatory cytokines. This study was planned to provide pharmacological basis for implementation of fresh-cut technology of Saposhnikoviae Radix from the producing area.
