[摘要]
目的 观察中药泽泻中指标成分23-乙酰泽泻醇B对细胞线粒体能量代谢的影响。方法 以SGC-7901胃癌细胞为研究对象,绘制其生长曲线,MTT法检测细胞增殖抑制率,以阳离子荧光羰花青染料JC-1染色,采用倒置荧光显微镜观察结合流式细胞仪检测对细胞线粒体膜电位;多功能酶标仪检测琥珀酸脱氢酶(SDH)活性。结果 23-乙酰泽泻醇B 3.125 μg•mL-1干预能促进细胞增殖,23-乙酰泽泻醇B 25 ~100 μg•mL-1干预则不同程度的抑制细胞增殖,并且呈浓度依赖关系。与正常对照组比较,不同剂量组的红色荧光和绿色荧光的变化不明显。流式细胞仪检测结果显示,正常对照组、23-乙酰泽泻醇B低、中、高剂量组的单阳细胞比例分别为3.5 %,3.6 %,3.7 %和4.0 %,与正常对照组比较,23-乙酰泽泻醇B低剂量组单阳细胞比例增加,但差异无统计学意义(P \> 0.05),23-乙酰泽泻醇B中、高剂量组单阳细胞比例增加,差异有统计学意义(P<0.05,P<0.01);SDH检测结果显示,正常对照组、23-乙酰泽泻醇B低、中、高剂量组的SDH活性分别为0.045、0.058、0.080、0.100 μmol•min-1,与正常对照组比较,23-乙酰泽泻醇B低剂量组的SDH活性差异无统计学意义(P \> 0.05),23-乙酰泽泻醇B中、高剂量组的SDH活性差异有统计学意义(P<0.05,P<0.01)。结论 23-乙酰泽泻醇B在低剂量时能促进SGC-7901细胞增殖,中剂量与高剂量则对SGC-7901线粒体膜电位有一定的损伤而抑制增殖,但对SDH活性促进作用较显著,提示其具有较明显促进线粒体能量代谢的作用,为进一步探讨泽泻改善线粒体能量代谢物质基础的研究奠定了基础。

[Abstract]
Objective To observe the effect of alisol B 23-acetate(ABA),an effective component of Rhizoma Alismae,on mitochondrial energy metabolism of SGC-7901 cells. Methods Human gastric carcinoma SGC-7901 cells were exposed to alisol B 23-acetate at different concentrations. The inhibitive rate of cell proliferation was measured by MTT assay and the cell growth curve was set up,SGC-7901 cells were dyed by cation fluorescence carbocyanine JC-1,the mitochondrial membrane potential was determined by the inverted fluorescence microscope and flow cytometer,and the succinate dehydrogenase(SDH) activity was detected by microplate reader. Results Alisol B 23-acetate at 3.125 μg/mL promoted cell proliferation,while alisol B 23-acetate at 25-100 μg/mL inhibited the proliferation of SGC-7901 cell line in a dose-dependent manner. Compared with the normal control group(NCG),the changes of red fluorescence weakening(or green fluorescence enhancing) in alisol B 23-acetate groups were not obvious. The proportion of single positive cells was 3.5 %,3.6 %,3.7 % and 4.0 % in NCG,and low-dose,medium-dose,and high-dose ABA groups,respectively,and the differences of NCG with medium- and high-dose ABA groups were significant(P<0.05,P<0.01). SDH activities of NCG,and low-dose,medium-dose,and high-dose ABA groups were 0.045,0.058,0.080,0.100 μmol•min-1,respectively,and the differences of NCG with medium- and high-dose ABA groups were significant(P<0.05,P<0.01). Conclusion Low dose of alisol B 23-acetate would promote SGC-7901 cell proliferation,while medium dose and high dose of alisol B 23-acetate would inhibit SGC-7901cells proliferation and accelerate SGC-7901cells SDH activity. It is implied that alisol B 23-acetate could improve the mitochondrial energy metabolism,which lay the foundation for investigating the therapeutic mechanism of Rhizoma Alismae on improving mitochondrial energy metabolism.
