[摘要]
目的 建立超高效液相色谱(UPLC)法同时测定肿节风超微粉中异嗪皮啶和迷迭香酸的含量,并比较不同粒度肿节风有效成分溶出率差异。方 法 采用UPLC法,Eclipse Plus C18(2.1 mm × 50 mm,1.8 μm),流动相为甲醇(A)-0.1 %磷酸(B),梯度洗脱,流速为0.4 mL·min-1,柱温为35 ℃,紫外检测波长 为342 nm。 结果 异嗪皮啶和迷迭香酸分别在9.97~59.84 μg·mL-1(r =0.99995),10.90~60.14 μg·mL-1(r=0.99935)范围内线性关系良好,平均回收率(n=6)分 别为95.16 %(RSD=2.15 %),100.31 %(RSD=2.29 %)。 结论 所建立的UPLC法操作简便快捷、经济、准确可靠、分离度好,可为肿节风药材有效成分分析和质量控制提供 新的方法。

[Abstract]
Objective To establish an ultra performance liquid chromatography(UPLC) method for the simultaneous determination of isofraxidin and rosmarinci acid in Herba Sarcandrae Glabrae,and to compare the dissolution rate of the active ingredients with different particle sizes. Methods UPLC was performed on Eclipse Plus C18(2.1×50 mm,RRHD 1.8 μm)with the mobile phase being methanol-0.1 % phosphoric acid by gradient elution. The flow rate was 0.4 mL·min-1,the column temperature was 35 ℃,and UV wave-length was 342 nm. Results Isofraxidin and marinci acid showed good linearity in 9.97 ~ 59.84 μg·mL-1(r =0.99995),10.90 ~ 60.14 μg·mL-1(r =0.99935),respectively. The mean recovery(n=6) was 95.16 %(RSD = 2.15 %),100.31 %(RSD = 2.29 %),respectively. Conclusion The established UPLC method is simple,rapid,economical,accurate and reliable with good separation for analysis of the active ingredients of Herba Sarcandrae Glabrae and its quality control.
