目的 观察β-细辛醚与冰片对β-淀粉样蛋白1-42(Aβ1-42)诱导PC12细胞损伤自噬相关蛋白轻链3(LC3)的作用,以及对细胞内钙离子浓度 和线粒体膜电位(MMP)的影响。方法 体外培养PC12细胞,β-细辛醚与冰片预处理,用终浓度为1.25 μmol·L-1 Aβ1-42诱导细胞产生自噬,光镜观察细胞形态,3-(4, 5-二甲基噻唑-2)-2,5-二苯基四氮唑溴盐比色法(MTT法)检测细胞活力,流式细胞仪检测LC3蛋白的表达、细胞内钙离子浓度及MMP,电镜观察细胞超微结构。结果 与模 型组比较,β-细辛醚组、冰片组及其β-细辛醚+冰片组能提高细胞活力(P< 0.05),升高LC3蛋白的含量(P< 0.05),降低细胞内钙离子浓度及升高MMP(P< 0.05), 减少自噬体,保护线粒体形态结构。结论 β-细辛醚与冰片及其配伍对Aβ1-42诱导的细胞损伤有保护作用。

[Abstract]
Objective To observe effects of β-asarone and borneol on autophagy-related protein light chain 3(LC3),calcium concentration and mitochondrial membrane potential(MMP) in PC12 cells with injury induced by amyloid β-protein(Aβ1-42). Methods PC12 cells were cultured in vitro and pretreated by β-asarone and borneol. Autophagy was induced by Aβ1-42 at the final concentration of 1.25 μmol·L-1. Cell morphology was observed under an inverted microscope. Cell viability was detected by methyl thiazolyl tetrazolium(MTT) assay. LC3 expression, intracellular calcium concentration and MMP were measured by flow cytometry. Ultrastructural morphology of PC12 cells was observed by transmission electron microscope. Results β-asarone,borneol and their combination increased cell viability and LC3 expression(P< 0.05),decreased intracellular calcium concentration(P< 0.05),increased MMP(P< 0.05),reduced autophagosome,and protected mitochondrial morphology. Conclusion β-asarone, borneol and their combination have protective effect on cell injury induced by Aβ1-42.
