[摘要]
目的 优化茜草中大叶茜草素、羟基茜草素的HPLC含量测定方法。方法 采用LUBEX Ecosil C18色谱柱(4.6 mm × 250 mm,5 μm),流动相为甲醇-乙腈-0.05 %磷酸水(25 ∶ 50 ∶ 25),流速1.0 mL·min-1,检测波长为250 nm。结果 大叶茜草素和羟基茜草素分别在19.34 ~ 161.20 μg·mL-1(r = 0.9999)和12.16 ~ 101.30 μg·mL-1(r = 0.9999)范围内呈良好线性,平均回收率分别为97.27 %(RSD=1.91 %)和100.00 %(RSD=1.87 %)。结论 该方法操作简单,重复性好,可作为茜草中大叶茜草素和羟基茜草素的含量测定方法,为合理制定茜草的含测方法提供参考。

[Abstract]
Objective To optimize the HPLC method for determining the content of rubimaillin and purpurin in Radix Rubiae. Methods LUBEX Ecosil C18 column(4.6 mm×250 mm,5 μm) was used with methanol- acetonitrile-0.05 % phosphoric acid solution as the mobile phase,the flow rate was 1.0 mL·min-1 and the PDA detective wavelength was set at 250 nm. Results The rubimaillin had a good linearity between 19.34~161.20 μg· mL-1(r=0.9999),the purpurin showed a good linearity within the range of 12.16~101.30 μg· mL-1(r=0.9999),and the recovery rates of rubimaillin and purpurin were 97.27 %(RSD=1.91%) and 100.00 %(RSD=1.87 %),respectively. Conclusion The method is simple,reproducible and stable,which can be used as the content determination method of rubimaillin and purpurin in Radix Rubiae.
