[摘要]
目的 完善溪黄草的定性定量方法,对药品质量进行有效控制。方法 采用薄层鉴别法,对溪黄草中迷迭香酸、齐墩果酸和芦丁进行鉴别。采用高效液相色谱(HPLC法)测定迷迭香酸的含量,以十八烷基硅烷键合硅胶为填充剂;以乙腈-0.1 %磷酸溶液(30∶70)为流动相;流速为1.0 mL·min-1;检测波长为330 nm;柱温为25 ℃;理论塔板数按迷迭香酸峰计算不低于4000。结果 TLC斑点清晰,分离度高,方法耐用性好;HPLC方法简便、专属性强,迷迭香酸线性范围为1.2 μg~12 μg(r = 0.9999),平均回收率为98.37 %,RSD为1.20 %(n=6)。结论 所建立的定性、定量方法可用于溪黄草的质量控制。

[Abstract]
Objective To optimize the qualitative and quantitative methods for Herba Rabdosiae Serrae,thus to improve its quality standard. Methods Thin layer chromatography(TLC) was performed for identifying rosmarinci acid,oleanolic acid and rutin. Rosmarinci acid was determined by high performance liquid chromatography(HPLC). The HPLC was conduced with octadecylsilane chemically bonded silica as loading agent,acetonitrile-0.1 % phosphoric acid solution(30∶70) as mobile phase,and the detection wavelength was 330 nm,flow rate was 1.0 mL·min-1 and column temperature was 25 ℃. The number of theoretical plates for rosmarinci acid was not less than 4000. Results The TLC spots were clear and can be well separated,indicating that the method had good ruggedness. The HPLC method was simple and specific. The calibration curve showed good linear relationship in the range of 1.2 μg~12 μg (r=0.9999). The average recovery was 98.37 % and RSD was 1.20 %(n=6). Conclusion The developed qualitative and quantitative methods can be used for the quality control of Herba Rabdosiae Serrae.
