[摘要]
目的 建立同时测定冠心Ⅴ号颗粒中丹参素和丹参酮ⅡA含量的HPLC方法。方法 采用高效液相色谱法,以甲醇-0.5 %醋酸水溶液梯度洗脱,色谱柱为Kromasil C18柱(4.6×250 mm,5 μm),流速为1.0 mL·min-1,检测波长为281 nm,柱温为35 ℃。结果 丹参素和丹参酮ⅡA分别在0.1~0.8 μg和0.02~0.16 μg范围内呈良好的线性关系;平均回收率分别为100.17 %和99.35 %。结论 本方法简便可行、重复性好,可用于同时测定冠心Ⅴ号颗粒中丹参素和丹参酮ⅡA的含量。

[Abstract]
Objective To establish a HPLC method for the simultaneous determination of danshensu and tanshinone ⅡA in Guanxin Ⅴ Granules by HPLC. Methods By using HPLC,the analysis was carried out on a column of Kromasil C18(4.6×250 mm,5 μm) with a gradient mobile phase of methanol and 0.5 % acetic acid at the flow rate of 1.0 mL·min-1 and column temperature of 35 ℃. The detection wavelength was 281nm. Results The linearity ranges of danshensu and tanshinone ⅡA were 0.1~0.8 μg and 0.02~0.16 μg,respectively,and the average recoveries were 100.17 and 99.35 %,respectively. Conclusion The established method is simple and reproducible,and can be used to simultaneously determine danshensu and tanshinone ⅡA in Guanxin Ⅴ Granules.
