目的 观察雷公藤内酯醇对佐剂性关节炎(adjuvant arthritis,AA)大鼠的抗炎作用。方法 采用完全弗氏佐剂复制AA大鼠模型,并选取模型复制成功的大鼠随机分为5组:即正常对照组,模型组,雷公藤内酯醇低、中、高剂量组,分别给予生理盐水和100,200,300 μg·kg-1的雷公藤内酯醇,灌胃给药14 d。采用酶联免疫吸附法(ELISA)检测大鼠白介素1β(IL-1β)、肿瘤坏死因子(TNF-α)及魏氏法检测血沉水平,观察踝关节病理变化。结果 与模型组比较,雷公藤内酯醇组各剂量组大鼠血清IL-1β、TNF-α和血沉水平明显降低(P < 0.05,P < 0.01),下降水平呈明显的剂量相关性,其中雷公藤内酯醇高剂量组(300 μg·kg-1)降低最显著。结论 雷公藤内酯醇对AA具有良好治疗作用,作用机制与其下调AA大鼠外周血中1L-1β、TNF-α水平有关。

[Abstract]
Objective To observe the anti-inflammatory effect of triptolide in adjuvant arthritis(AA) rats. Methods AA model was induced with Freund's complete adjuvant,and then the rats presenting secondary response were randomly assigned into model group,triptolide groups(triptolide in the dosage of 100,200,300 μg·kg-1,respectively). Meanwhile,eight normal rats were randomly selected as the normal control group,and were given intragastric administration of normal saline. Rats in the medication groups were given correspondent drugs by intragastric administration. After administration for 14 days,the serum levels of interleukin 1β(IL-1β) and tumor necrosis factor alpha(TNF-α) were measured by enzyme-linked immunosorbent assay(ELISA),and the erythrocyte sedimentation rate(ESR) was measured. The pathological changes of ankle joint were also observed. Results In comparison with the model group,serum levels of IL-1β,TNF-α and ESR in triptolide groups were significantly decreased(P < 0.05,P < 0.01),and the decrease in high-dose triptolide group was more obvious(P < 0.01). Conclusion Triptolide shows significantly therapeutic effect on AA in rats,and its therapeutic mechanism may be related with the down-regulation of serum levels of IL-1β and TNF-α in AA rats.
