[摘要]
目的 建立测定柴葛解肌汤中Ⅰ型柴胡皂苷a与Ⅱ型柴胡皂苷b2的HPLC-MS/MS方法。方法 采用Hypersil GOLD aQ色谱柱(150 mm×2.1 mm,3 μm),乙腈-0.1 %甲酸溶液为流动相,流速为300 μL·min-1,柱温:25 ℃,以液相色谱分离、电喷雾离子化串联进行检测。结果 柴胡皂苷a、b2分别在44.8~672 ng·mL-1、47.2~708 ng·mL-1线性关系良好(r=0.9992、r=0.9993),平均回收率分别为98.53 %和99.47 %,RSD分别为2.01 %和2.08 %(n=6)。结论 该方法快速简便、准确、灵敏度高,可用于柴葛解肌汤中柴胡皂苷a、b2的含量测定。

[Abstract]
Objective To establish an HPLC-MS /MS method for the determination of saikosaponin a and saikosaponin b2 in Chaige Jieji decoction. Methods The separation was performed by using HPLC method with Hypersil GOLD aQ(150 mm ×2.1 mm,3 μm)as chromatographic column. The mobile phase consisted of acetonitrile -0.1 % formic acid and the flow rate was 300 μL·min-1, the column temperature was set at 25 ℃,and the detection was done by electrospray ionization mass spectrometry. Results The linear range was 44.8~672 ng·mL-1 for saikosaponin a(r=0.9992),and 47.2~708 ng·mL-1 for saikosaponin b2(r=0.9993). The average recovery rates of saikosaponin a and saikosaponin b2 were 98.53 %,99.47 %,and RSD were 2.01 %,2.08 %(n=6),respectively.Conclusion The established method is simple,quick,sensitive and accurate,can be used to determine the content of saikosaponin a and saikosaponin b2 in Chaige Jieji decoction.
