目的 观察疏风解毒胶囊体内抗呼吸道合胞病毒(RSV)、单纯疱疹病毒I型(HSV-I)和柯萨奇病毒B3型(CVB3)的作用。方法 分别用RSV、HSV-I、CVB3感染小鼠建立模型,实验分疏风解毒胶囊高、中、低剂量组,利巴韦林组,清开灵组,病毒对照组及正常对照组,灌胃给药,记录各组小鼠死亡数及存活时间。以肺指数、动物死亡保护率及生命延长率等指标观察疏风解毒胶囊体内抗病毒的作用。结果 与病毒对照组比较,疏风解毒胶囊能明显降低RSV感染小鼠肺指数,并存在剂量依赖关系,其肺指数抑制率略低于利巴韦林组,而明显高于清开灵组。同时疏风解毒胶囊对3种病毒所致的动物死亡保护率、生命延长率等指标均明显增加,与上述对肺指数抑制率的结果吻合。结论 疏风解毒胶囊可有效地改善RSV感染所引起的小鼠肺部肿胀,对RSV、HSV-I、CVB3病毒感染小鼠均有明显保护作用。

[Abstract]
objective To observe the anti-viral effects of Shufeng Jiedu capsule on mice models infected with respiratory syncytial virus(RSV),herpes simplex virus type I(HSV-I) and coxsackie virus B3 type(CVB3)respectively. Methods Mice models of RSV,HSV-I or CVB3 infection were established. High dose,moderate dose and low dose of Shufeng Jiedu capsule were administrated intragastrically to the model mice,and Ribavirin and Qingkailing granula were used as positive control medicine. The uninfected mice served as normal control. Mice survival time and the number of surviving mice were recorded,and the antiviral activity of Shufeng Jiedu capsule was evaluated by lung index,the rate of death protection and lifespan extension. Results In comparison with the viral infection model group,Shufeng Jiedu capsule significantly reduced the increase of lung index of mice with RSV infection in a dose-dependent manner,and its inhibitory rate for lung index was slightly lower than that of Ribavirin particles,but significantly higher than that of Qingkailing particles. The death protection rate and lifespan extension rate of three kinds of mice model were increased after treatment with Shufeng Jiedu capsule,and the results were in accordance with the inhibitory rate for lung index. Conclusion Shufeng Jiedu capsule can significantly relieve the pulmonary edema in RSV-infected mice,and has protective effect on mice infected with RSV,HSV-I,CVB3.

安徽省自然科学基金项目资助(1308085MH129)。 中为了模拟自然感染,全部采用滴鼻感染的方式,在LD50的测定时发现3种病毒滴鼻感染均可达到小鼠感染致死的预期目的。在病毒的感染剂量上,本实验根据预实验的结果进行了一些调整,RSV病毒液增加到0.5 mL。与以往腹腔注射植入病毒的方法比较[2,4-6],滴鼻法更加接近自然环境下感染,避免了腹腔感染或腹部创伤对实验结果的影响。但滴鼻法所产生的非特异性炎症以及实验中可能导致实验动物窒息,对实验结果可能产生影响。故在以后的实验中,将尽可能地将病毒的毒力增强,减少感染病毒液的剂量,从而降低滴鼻所导致的非特异性炎症对实验结果的影响。在小鼠药物剂量设计时,我们参照刘颖等[1]方法进行,剂量略高于临床实际用量。在以后的实验里,也宜接近临床剂量,为临床提供更准确的实验依据。
