[摘要]
目的 观察合欢花总黄酮对抑郁模型大鼠海马CA1区脑源性神经营养因子(BDNF)及其受体酪氨酸激酶B(TrkB)表达的影响。方法 将90只SD大鼠随机分为正常对照组,模型组,盐酸文拉法辛组(12.5 mg·kg-1),合欢花总黄酮高、中、低剂量组(100,50,25 mg·kg-1)。以孤养加慢性不可预见性应激方法复制大鼠抑郁模型。采用旷场实验法测定各组大鼠的行为变化,免疫组织化法检测大鼠海马CA1区BDNF及TrkB表达。结果 与正常对照组比较,模型组大鼠旷场实验3 min内水平运动、垂直运动得分均明显减少(P < 0.05,P < 0.01),海马CA1区BDNF及受体TrkB的表达降低(P < 0.01);与模型组比较,各给药组水平运动得分和垂直运动得分都明显增加(P < 0.05);海马CA1区BDNF及受体TrkB的表达明显增加(P < 0.05,P < 0.01)。结论 合欢花总黄酮可以改善抑郁模型大鼠的抑郁行为,具有抗抑郁功效,其作用机制可能与增加海马CA1区BDNF及其受体TrkB的表达,从而保护海马神经元有关。

[Abstract]
Objective To observe the effect of Flos Albizziae total flavonoids(FATF) on the expression of brain-derived neurotrophic factor(BDNF) and its receptor tyrosine kinase B(TrkB) in the hippocampus CA1 of rats with depression. Methods Ninety SD rats were randomly divided into normal group,model group,Venlafaxine Hydrochloride group(12.5 mg·kg-1),and high-,middle-,low-dosage FATF groups(100,50,25 mg·kg-1,respectively). Depression rat model was established by solitary cultivation plus unpredictable chronic stress. Rats in Venlafaxine Hydrochloride group and FATF groups were orally administered with Venlafaxine Hydrochloride or FATF once daily for 21 days,and rats in normal group and model group were given equal volume of distilled water. The behavior scores were evaluated by open field test,and the expression of BDNF and Trkb in hippocampal CA1 was detected by immunohistochemical assay. Results Compared with the normal group,the behavior scores in the model group were decreased significantly(P < 0.05 or P < 0.01),and the expression of BDNF and TrkB in the hippocampal CA1 was significantly decreased(P < 0.01). Compared with the model group,the behavior scores were increased(P < 0.05) in Venlafaxine Hydrochloride group and FATF groups,and the expression of BDNF and TrkB in the hippocampal CA1 was significantly increased(P < 0.05 or P < 0.01). Conclusion FATF can improve the behavior of rats with depression,having antidepressant effect,and its mechanism in protecting hippocampus neurons may be related to the improvement of the expression of BDNF and TrkB in the hippocampal CA1.
