[摘要]
目的 建立HPLC-ELSD法测定更年调和颗粒中黄芪甲苷的含量。方法 色谱柱为Thermo Syncronis C18柱(250 mm×4.6 mm,5 μm);流动相为乙腈-水(34 ∶ 66);流速:1.0 mL·min-1;柱温:30 ℃。蒸发光散射检测器(ELSD)参数:漂移管温度85 ℃,氮气体积流量1.8 mL·min-1。结果 黄芪甲苷在0.812~8.12 μg范围内具有良好的线性关系,加样回收率为98.07 %(RSD为1.21 %,n=6)。结论 本法简单易行,重复性符合要求,测定结果准确,可作为更年调和颗粒中黄芪甲苷的含量测定方法。

[Abstract]
Objective To establish a HPLC-ELSD method for the determination of astragaloside IV in Gengnian Tiaohe Granula. Methods The HPLC-ELSD system consisted of Thermo Syncronis C18 column(250 mm × 4.6 mm,5 μm) with the mobile phase of acetonitrile-water(34 ∶ 66). The flow rate was 1.0 mL·min-1,and the column temperature was set at 30 ℃. Evaporative light-scattering detector was adopted. The drift tube temperature of ELSD was set at 85 ℃,nitrogen volume flow rate was 1.8 L·min-1. Result Astragaloside IV showed a good linearity in the range of 0.812~8.12 μg. The recovery was 98.07 %(RSD=1.21 %,n=6). Conclusion The established method is simple and reliable,and is suitable for the determination of astragaloside IV in Gengnian Tiaohe Granula.
