[摘要]
目的 探讨奥瑞凝胶治疗反流性食管炎的作用机制。方法 采用半幽门结扎加贲门肌切开术复制反流性食管炎大鼠模型。大鼠随机分为假手术组,赋形剂组,奥瑞凝胶高、中、低剂量组,雷尼替丁组,模型复制术后7 d开始给药,连续给药14 d后,观察各组大鼠食管黏膜一氧化氮合酶(NOS)及血管活性肠肽(VIP)的表达。结果 奥瑞凝胶各剂量组与赋形剂组比较,大鼠食管黏膜NOS和VIP表达明显下调(P < 0.01)。结论 奥瑞凝胶通过影响食管黏膜组织NOS和VIP的分泌以提高下食管括约肌的压力,减少一过性下食管括约肌松弛,从而达到治疗反流性食管炎的目的。

[Abstract]
Objective To explore the therapeutic mechanism of Aorui Gel for the treatment of reflux esophagitis(RE). Methods The SD rats were randomly divided into 6 groups,sham operation group,excipient gel Carbomer group,ranitidine positive control group,and high-,medium- and low-dose Aorui Gel treatment group. Reflux esophagitis rat model was established by semi-pyloric ligation & cardiac muscle incision methods. Seven days after the successful preparation of the model,the medication started and continued for 14 days. After treatment,the expression of nitric oxide synthase(NOS) and vasoactive intestinal polypeptide(VIP) in the esophagus tissue was examined by immunohistochemical method. Results The average gray value of the NOS and VIP positive expression in esophageal mucosa was down-regulated in Aorui Gel groups,and the differences were significant when compared with the Carbomer group(P < 0.01). Conclusion Aorui Gel can down-regulate the esophageal NOS and VIP levels to regulate the lower esophageal sphincter,so as to achieve the purpose of treating RE.
