[摘要]
目的 比较侧柏叶药材及其水煎液高效液相色谱(HPLC)特征图谱的差异,探讨其特征图谱的相关性。方法 采用Zorbax SB-Aq C18色谱柱;流动相为乙腈-磷酸溶液(体积分数为0.1 %),梯度洗脱;检测波长为254 nm;柱温为40 ℃;流速为1.0 mL·min-1。结果 侧柏叶药材标示出22个共有特征峰,水煎液标示出药材的16个特征峰,鉴别了5个黄酮类特征峰(杨梅苷、异槲皮苷、槲皮苷、槲皮素、穗花杉双黄酮)。分别以10批药材及其水煎液特征图谱生成的共有模式为对照,各批次药材及其水煎液的相似度分别大于0.99和0.90。同批次药材及其水煎液特征图谱之间的相似度,有7批大于0.95,3批低于0.90。结论 水煎煮方式能大部分提取侧柏叶的主要特征成分。本研究为侧柏叶粉末及含侧柏叶复方粉末水煎液外用的物质基础研究提供了一定的依据。

[Abstract]
Objective To compare the differences of characteristic spectrum for Cacumen Platycladi and its decoction by high pressure liquid chromatography(HPLC) and to explore the correlation of characteristic spectrum. Methods HPLC was performed on the Zorbax SB-Aq C18 column,the acetonitrile-phosphoric acid solution(volume fraction being 0.1 %) by gradient elution was employed as the mobile phase,detection wavelength was set at 254 nm,column temperature was 40 ℃,and flow rate was 1.0 mL·min-1.Results Twenty-two common peaks were shown in the medicinal powder of Cacumen Platycladi and 16 were separated in the decoction of Cacumen Platycladi. Five peaks were identified as myricitrin,isoquercitrin,quercitrin,quercetin and amentoflavone. The similarities of 10 batches of the medicinal powder and their decoction were over 0.99 and 0.90,respectively. The correlation coefficient of 7 batches between them was over 0.95,and that of the other 3 batches was below 0.90. Conclusion The decocting method is effective for obtaining the main characteristic components of Cacumen Platycladi.The study can provide a reference for the external application of Cacumen Platycladi powder and its compound preparation powder.
