目的 研究苦瓜水提物(Momordica Charantia Aqueous Extract, MCAE)对环磷酰胺(Cyclophosphamide,Cy)致免疫抑制小鼠的肠道黏膜免疫功能的影响。方法 通过CCK-8法检测肠系膜淋巴结(mesenteric lymph nodes,MLN)和派氏结(Peyer’s Patches,PPs)淋巴细胞转化增殖;流式细胞术检测MLN及PPs中B淋巴细胞的比例和活化水平;ELISA试剂盒检测肠道sIgA的分泌水平。结果 与Cy诱导的免疫抑制模型组比较,MCAE中、高剂量(200,400 mg·kg-1)分别对Cy抑制的小鼠MLN和PPs淋巴细胞转化增殖能力有一定的上调作用(P < 0.05);MCAE中剂量对MLN和PPs中B淋巴细胞比例及活化水平的降低同样具有一定的上调作用(P < 0.05,P < 0.01)。结论 苦瓜水提物对Cy诱导的肠道黏膜免疫功能低下有一定的上调作用,其作用机制可能与MLN和PPs中B淋巴细胞的增殖、活化有关。

[Abstract]
Objective To study the effects of Momordica charantia aqueous extracts(MCAE) on intestinal mucosa in immunosuppressive mice induced by cyclophosphamide. Methods Cell proliferation was detected by CCK-8 in mesenteric lymph nodes(MLN) and the Peyer’s Patches(PPs),and the proportion and activation of B lymphocytes were detected by flow cytometry in MLN and PPs. Intestinal sIgA was detected by ELISA. Results Compared with the immunosuppressive group,the groups of middle dose of and high dose of MACE significantly up the regulated the dropping ability of proliferation of MLN and PPs lymphocytes. Middle dose of MACE could increase the proportion level of B cells in MLN and PPs. Conclusion Momordica charantia aqueous extracts can up-regulalate intestinal mucosa immune in immunosuppressive mice induced by cyclophosphamide,and its mechanism is closely related to the proliferation and activation of B lymphocytes in MLN and PPs.
