[摘要]
目的 观察疏风解毒胶囊体外抑制病毒及细菌的作用。方法 采用细胞病变(CPE)抑制法,以利巴韦林颗粒和清开灵颗粒为对照,从药物的最大无毒浓度开始,测定疏风解毒胶囊对呼吸道合胞病毒(respiratory syncytial virus,RSV)、柯萨奇病毒B3型(coxsackie virus B3,CVB3)和单纯疱疹病毒1型(herpes simplex virus1,HSV-1)的抑制作用。按Reed-muench法计算50 %细胞毒性浓度(TC50),最大无毒浓度(TC0),50 %抑制浓度(IC50)和药物治疗指数(TI)。采用平皿稀释法及抑菌环试验,以头孢曲松钠注射液、清开灵颗粒为对照,检测疏风解毒胶囊对金黄色葡萄球菌、大肠埃希菌、铜绿假单胞菌、志贺氏痢疾杆菌、肺炎链球菌、乙型链球菌的最小抑菌浓度(MIC)和最小杀菌浓度(MBC)。结果 疏风解毒胶囊的TC50为2-0.3 /mL,TC0为2-0.7 / mL;利巴韦林分别为2-0.4 / mL和2-1.7 / mL,清开灵分别为20.7 / mL和2-1.7 / mL。疏风解毒胶囊对RSV的IC50为2-8 / mL,TI为27.7;利巴韦林分别为2-5.7 / mL和25.4,清开灵分别为2-9.4 / mL和29.1;疏风解毒胶囊对CVB3的IC50为2-5.4 / mL,TI为25.1,利巴韦林分别为2-4.3 / mL和24,清开灵分别为2-8.3 / mL和28;疏风解毒胶囊对HSV-1的IC50为2-6.8 / mL,TI为26.5,利巴韦林分别为2-5.3 / mL和25,清开灵分别为2-8.3 / mL和28。疏风解毒胶囊对金黄色葡萄球菌,大肠埃希菌,铜绿假单胞菌,志贺氏痢疾杆菌,肺炎链球菌和乙型链球菌的MIC和MBC分别为0.1, 0.05,1.62,0.8,0.8,0.4和1.62,0.8,0.025,< 0.025,0.2,0.1 mg·mL-1;头孢曲松钠注射液对上述6种细菌的MIC均为0.125 μg·mL-1,MBC均 < 0.125 μg·mL-1;而清开灵颗粒对以上6种细菌均无抑菌作用。抑菌环实验证明疏风解毒胶囊对6种细菌的抑制作用弱于对照药头孢曲松钠,而清开灵颗粒未见其抑菌作用。结论 疏风解毒胶囊对多种病毒和细菌都有一定的抑制作用,其抗病毒作用弱于利巴韦林,但其细胞毒作用较利巴韦林低;其抗病毒和抗菌作用优于清开灵颗粒。

[Abstract]
Objective To study the antiviral and antibacterial effects of Shufeng Jiedu capsules(SJC) in vitro. Methods The antiviral effect of SJC on respiratory syncytial virus(RSV),coxsackie virus B3(CVB3) and herpes simplex virus 1(HSV-1) was investigated by the cytopathic effect(CPE) inhibitory assay in Hep-2 cell culture,with medicines of ribavirin and Qingkailing granula as the positive control. According to Reed-muench assay,cell cytotoxic concentration of 50 percentage(TC50),maximum non-toxic concentration(TC0),inhibitory concentration of 50 percentage (IC50) and therapeutic index(TI) were calculated. In addition,with ceftriaxone sodium injcetion and Qingkailing granula as the positive control,the antibacterial effects of SJC on Staphylococcus aureus,Escherichia coli,Pseudomonas aeruginosa,Shigella dysenteriae,Streptococcus pneumoniae,and β-Hemolytic streptococcus were observed by detecting the minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC) by plate dilution method and the antibacterial zone diameters test. Results For SJC,TC50 was 2-0.3/mL,and TC0 was 2-0.7/mL. For ribavirin,TC50 and TC0 were 2-0.4/mL and 2-1.7/mL,respectively. Qingkailing had TC50 of 20.7/mL and TC0 of 2-1.7/mL. IC50 and TI of SJC,ribavirin and Qingkailing to100 TCID50 of RSV strain infection were to 2-8/mL and 27.7, 2-5.7/mL and 25.4, 2-9.4mL and 29.1,respectively. IC50 and TI of SJC,ribavirin and Qingkailing to 100TCID50 of CVB3 strain infection were 2-5.4/mL and 25.1,2-4.3/mL and 24,2-8.3/mL and 28,respectively. To 100 TCID50 of HSV-1 strain infection,IC50 and TI of SJC,ribavirin and Qingkailing were 2-6.8/mL and 26.5,2-5.3/mL and 25,2-8.3/mL and 28,respectively. The resutls of antibacterial test showed that MIC of SJC on Staphylococcus aureus,Escherichia coli,Pseudomonas aeruginosa,Shigella dysenteriae,Streptococcus pneumoniae and β-Hemolytic streptococcus was 0.1,1.62,0.8,1.62, 0.025 and 0.2 mg·mL-1,respectively,and MBC was 0.05,0.8,0.4,0.8,less than 0.025 and 0.1 mg·mL-1,respectively. MIC and MBC of ceftriaxone sodium injection for the above six kinds of bacteria were 0.125 μg·mL-1 and less than 0.125 μg·mL-1,respectively. Qingkailing had no effect on the above six kinds of bacteria. The results of antibacterial zone diameters test indicated that the effects of SJC on six kinds of bacteria were weaker than those of ceftriaxone sodium,while Qingkailing had no antibacterial effect on them. Conclusion SJC show certain inhibition on various viruses and bacteria in vitro,its antiviral effect is slightly weaker than ribavirin,but its cytotoxic effect is lower than ribavirin. Its antiviral and antibacterial effects are much better than Qingkailing granula.
