[摘要]
目的 建立同时测定三黄颗粒中黄芩苷、小檗碱含量的HPLC法。方法 采用Kromasil C18(4.6 mm × 250 mm,5 μm)色谱柱为固定相,以甲醇-0.1 %磷酸水溶液为流动相,梯度洗脱,流速:1.0 mL·min-1,柱温:25 ℃,进样量:10 μL,检测波长:黄芩苷为280 nm,盐酸小檗碱为345 nm。结果 黄芩苷、小檗碱的线性范围分别为0.2048~2.0480 μg(r = 0.9999),0.2160~2.160 μg(r = 0.9999);平均加样回收率分别为99.89 %、99.98 %,RSD分别为1.45 %、1.76 %。结论 本方法操作简便快捷,结果准确可靠,重复性好,可用于三黄颗粒的质量控制。

[Abstract]
Objective To establish a HPLC method for the simultaneous determination of baicalein and berberin in Sanhuang Granules. Methods The HPLC analysis was performed on a Kromasil C18 column(250mm×4.60 mm,5 μm)with methanol-water(0.1 % phosphonic acid) as mobile phase in gradient elution. The flow rate was set at 1.0 mL·min-1 and the ultraviolet detector was operated at 280 nm for Baicalein and 345 nm for Berberin,respectively. Results Baicalein and berberin showed good linearity in the range of 0.2048~2.0480 μg(r=0.9999)and 0.2160~2.160 μg(r = 0.9999) respectively. The average recoveries of adding sample were 99.89 % and 99.98 %,respectively and RSD(n = 6) was 1.45 %,and 1.76 %,respectively. Conclusion The established method is simple,accurate and reproducible,which is suitable for the determination of baicalein and berberin in Sanhuang Granules.
