目的 观察不同浓度的灵芪蠲肝胶囊含药血清对血小板衍化生长因子(PDGF)诱导的大鼠肝星状细胞-T6(HSC-T6)增殖和组织金属蛋白酶抑制剂-1(TIMP-1)蛋白量表达的影响，探讨其可能的抗肝纤维化机制。方法 灵芪蠲肝胶囊高、中、低剂量组按含药血清浓度200 mL·L-1分别作用于10 ng·mL-1 PDGF刺激的HSC-T6，24 h、48 h、72 h后，用CCK-8法测定各组HSC-T6的吸光度值，透射电镜观察各组细胞超微结构的变化，免疫印迹法检测24 h时各组细胞中TIMP-1蛋白量的表达。结果 灵芪蠲肝胶囊含药血清可明显抑制HSC-T6的增殖，各组药物在各时间点对HSC-T6的抑制作用随血清中药物浓度增加呈逐渐增强趋势；同时各药物血清组HSC-T6中TIMP-1蛋白表达量均明显减少(均P < 0.01)。结论 (1)灵芪蠲肝胶囊可以抑制PDGF诱导的HSC-T6增殖，且作用具有剂量依赖性；(2)灵芪蠲肝胶囊能降低TIMP-1在活化的HSC-T6中的表达，从而减少其对基质金属蛋白酶(MMPs)的抑制，实现抗肝纤维化作用。

Ojective To investigate the effects of serum containing Lingqi Juangan Capsule(LJC) at different concentrations on the proliferation of hepatic stellate cell-T6(HSC-T6) and the expression of tissue inhibitor of metalloproteinase-1(TIMP-1) proteins in HSC-T6 induced by platelet-derived growth factor(PDGF). Methods The drug-containing serum was obtained by intragastric administration of high-，middle- and low-dose LJC decoction to the SD rats respectively. HSC-T6 induced by 10 ng·mL-1 PDGF was incubated at 200 mL·L-1 of the drug serum containing different doses of LJC. After 24，48，and 72 hours，the proliferation of HSC-T6 was measured by cell counting Kit-8(CCK-8) and the growth curve was made. The ultrastructure of HSC-T6 was observed under transmission electron microscope. After 24 hours，the expression of TIMP-1 of HSC-T6 in each group was measured by Western blotting. Results LJC-containing serum had an obvious effect on inhibiting the proliferation of HSC-T6，and the effect was dose-dependant at each time point. The expression of TIMP-1 was increased in each drug-containing serum group(P < 0.01). Conclusion LJC can inhibit the proliferation of HSC-T6 in a dose-dependent manner，and its anti-hepatic fibrosis mechanism is related with the decrease of TIMP-1 expression in activated HSC，and with alleviating the inhibition to MMPs.
