[摘要]
目的 建立地龙氨基酸类成分含量测定方法,评价不同产地的地龙质量。方法 采用高效液相色谱法测 定 24 批不同产地地龙中氨基酸类成分含量,并采用主成分分析(PCA)及偏最小二乘法判别分析(PLS-DA)对地 龙质量进行评价。结果 地龙中天冬氨酸、谷氨酸、丝氨酸、组氨酸、甘氨酸、精氨酸、脯氨酸、丙氨酸、缬 氨酸、甲硫氨酸、异亮氨酸、亮氨酸、色氨酸、苯丙氨酸、赖氨酸、酪氨酸在各自范围内线性关系良好(r> 0.999),平均加样回收率为 98.73%~102.56%,RSD 在 1.93%~3.18% 范围内,方法学验证符合 《中国药典》 含量测定的要求。PCA 分析提取到 3 个主成分,累积方差贡献率为 82.493%,其中两广地区产地龙综合得分较 高,排名靠前。PLS-DA 分析提示,脯氨酸、甲硫氨酸、组氨酸、丙氨酸、苯丙氨酸和色氨酸是影响地龙质量 的主要差异性标志物。结论 基于氨基酸类成分定量结合化学计量学的评价方法,可以用于不同产地的地龙质 量差异研究。

[Abstract]
Objective To establish a method for determining the content of amino acid components in Pheretima and to evaluate the quality of pheretima from different origins. Methods The contents of amino acid components in 24 batches of Pheretima from different origins were determined by high-performance liquid chromatography (HPLC). Principal component analysis (PCA) and partial least squares-discriminant analysis (PLS-DA) were used to evaluate the quality of Pheretima. Results Aspartic acid,glutamic acid,serine,histidine,glycine,arginine,proline,alanine, valine, methionine, isoleucine, leucine, tryptophan, phenylalanine, lysine, and tyrosine showed good linear relationships within their respective ranges (r>0.999). The average recovery rates ranged from 98.73% to 102.56%,with RSD values ranging from 1.93% to 3.18%,meeting the requirements of the Chinese Pharmacopoeia for content determination. PCA extracted three principal components with a cumulative variance contribution rate of 82.493%. Pheretima samples from the Guangdong and Guangxi regions had higher comprehensive scores and ranked at the top. PLS-DA indicated that proline, methionine, histidine, alanine, phenylalanine, and tryptophan were the main differential markers affecting the quality of Pheretima. Conclusion The evaluation method based on quantitative analysis of amino acid components combined with chemometrics can be used to study the quality differences of Pheretima from different origins.
