[摘要]
目的 基于高效液相色谱法(HPLC)建立同时测定桂郁金 7 种成分(莪术二醇、去甲氧基姜黄素、姜黄 素、莪术烯醇、异莪术烯醇、吉马酮、呋喃二烯)含量的方法。方法 采用 HPLC 法,Waters Symmetry C18色谱 柱(4.6 mm×250 mm,5 µm),以乙腈(A)-0.2% 磷酸水溶液(B)为流动相梯度洗脱;流速 0.8、0.8~1.0 mL·min-1 , 检测波长 204 nm,柱温 30 ℃,进样量 10 µL。结果 桂郁金 HPLC 图谱中 7 种成分的分离度良好。莪术二醇、 去甲氧基姜黄素、姜黄素、莪术烯醇、异莪术烯醇、吉马酮、呋喃二烯的线性范围分别为 4.84~48.36、0.08~ 3.36、0.16~3.25、3.20~32.00、0.32~3.22、1.61~32.24、0.32~6.48 µg·mL-1 ,各成分线性关系良好(r≥0.999 5)。 7 种成分的平均加样回收率为 96.37%~108.47%,RSD 值≤2.14%。上述 7 种成分在 22 批桂郁金样品中的平均 含量分别为 308.39、14.88、25.30、328.72、21.83、152.00、46.20 µg·g-1 。结论 本方法准确、可靠,可用于 桂郁金的质量评价及其 7 种成分的含量测定,可为桂郁金的质量控制提供科学依据。

[Abstract]
Objective To establish a method for the simultaneous determination of seven components (aerugidol, demethoxycurcumin, curcumin, curcumenol, isocurcumenol, germacrone, and furanodiene) in Curcumae Radix based on high performance liquid chromatography (HPLC). Methods HPLC analysis was performed using a Waters Symmetry C18 column (4.6 mm × 250 mm,5 µm). A gradient elution system consisting of acetonitrile (A) and-0.2% phosphoric acid aqueous solution (B) was used as the mobile phase. The flow rate was 0.8,0.8-1.0 mL·min-1 ,the detection wavelength was set at 204 nm,the column temperature was maintained at 30 ℃,and the injection volume was 10 µL. Results The seven components in Curcumae Radix were well separated. The linear ranges for aerugidol, demethoxycurcumin,curcumin,curcumenol,isocurcumenol,germacrone,and furanodiene were 4.84-48.36,0.08- 3.36, 0.16-3.25, 3.20-32.00, 0.32-3.22, 1.61-32.24, and 0.32-6.48 µg·mL-1 , respectively. All components exhibited excellent linear relationships (r ≥ 0.999 5). The average recovery rates ranged from 96.37% to 108.47%,with RSD ≤ 2.14%. In the analysis of 22 batches of Curcumae Radix samples,the average contents of the aforementioned seven components were 308.39,14.88,25.30,328.72,21.83,152.00,and 46.20µg·g-1 ,respectively. Conclusion This method is accurate and reliable, suitable for the quality evaluation of Curcumae Radix and the quantitative determination of its seven characteristic components. It can provide a scientific basis for the quality control of Curcumae Radix.
